چکیده

To explore the association of serum hyperhomocysteinaemia with 677C>T polymorphism in MTHFR gene among metformin-treated patients with type 2 diabetes. Serum-homocysteine levels of 227 diabetic subjects were measured before and after the metformin administration were analyzed statistically with respect to their genotype data for MTHFR 677C>T polymorphism. CC was the most prevalent genotype at MTHFR 677C>T in our sample and associated with the lowest homocysteine level. The patients with TT genotype had significantly elevated homocysteine compared with CC following metformin use (ORBaseline = 3.434 and 95% CI:0.622–4.127 vs. OREndpoint = 4.466 95% CI:3.124–5.53). A distinct statistical association between hyperhomocysteinaemia and the carriage of MTHFR 677C>T polymorphism was established among Indian diabetic patients who were treated with metformin. Our study underscores the importance of phamacogenetic analysis in diabetes-related therapeutics. Subjects: Endocrinology; Genetics; Statistics for the Biological Sciences

MTHFR C677T and A1298C gene polymorphisms and hyperhomocysteinemia as risk factors of diabetic nephropathy in type 2 diabetes patients.

Point mutations in methylenetetrahydrofolate reductase (MTHFR) and hyperhomocysteinemia were implicated in the pathogenesis of diabetic nephropathy (DN) in many ethnic groups. This study addressed the association of C677T and A1298C single nucleotide polymorphisms (SNPs) of MTHFR gene with DN in Tunisian type 2 diabetes (T2DM) patients. Study subjects comprised 93 DN patients, 267 patients with...

Methylene Tetrahydrofolate Reductase Gene Polymorphism and the Risk of Ischemic Stroke in Type 2 Diabetic Egyptian Patients

Background: A common C677T mutation of methylenetetrahydrofolate reductase (MTHFR) gene is reported to be associated with hyperhomocysteinemia a risk for atherosclerotic vascular diseases and ischemic stroke (IS). Aim: To clarify the possible role of the MTHFR 677C>T polymorphism in predisposition to IS and to assess the relationship of allelic variants of MTHFR gene with homocysteine (Hcy) pla...

Association of Methylenetetrahydrofolate Reductase (MTHFR 677C>T and 1298A>C) Polymorphisms and Haplotypes with Silent Brain Infarction and Homocysteine Levels in a Korean Population

PURPOSE Methylenetetrahydrofolate reductase (MTHFR) is the main regulatory enzyme for homocysteine metabolism. In the present study, we evaluated whether the MTHFR 677C>T and 1298A>C gene polymorphisms are associated with SBI and plasma homocysteine concentration in a Korean population. MATERIALS AND METHODS We enrolled 264 patients with SBI and 234 healthy controls in South Korea. Fasting pl...

Polymorphisms of methylenetetrahydrofolate reductase gene as the genetic predispositions of coronary artery diseases in eastern India

BACKGROUND Gene-environment interaction is an important aspect in the development of coronary artery disease (CAD). The mutation (677C-T) of methylenetetrahydrofolate reductase (MTHFR) gene results in a decrease of the enzyme activity that leads to mild hyperhomocysteinemia. Elevated plasma level of homocysteine has been recognized as an independent risk factor for cardiovascular disease. A cas...

Methylenetetrahydrofolate reductase polymorphism 677C>T is associated with peripheral arterial disease in type 2 diabetes

BACKGROUND Individuals with diabetes are twice as likely to develop peripheral arterial disease (PAD), the manifestation of extensive atherosclerosis throughout the lower extremities. One putative determinant of PAD is the 677C>T polymorphism in the gene encoding methylenetetrahydrofolate reductase (MTHFR), which has previously been found to associate with various diabetic complications includi...
